The Premier League club have made the 21-year-old Englishman a top target as they look to recruit a right-sided wing-back this summer.
A recruitment source has told Football Insider that Spurs have already been in contact over a deal for Spence as they look to beat Premier League rivals to his signature.
Woolwich are also keen on the wide man, who has made such a big impression while on loan at Nottingham Forest this season.
It is believed a fee of around £10million will be enough to complete a deal for Spence, the source added.
Boro are expected to cash in on the defender, who joined from the Fulham youth set-up in the summer of 2018, as he looks to take the next step in his career.
He has a contract at the Riverside Stadium that runs until 2024.
Spence has burnished his reputation this season while playing an instrumental role in Forest’s promotion bid.
He has made 40 league starts among 49 appearances in all competitions, and started both of their play-off semi-final wins over Sheffield United.
The attacking wing-back is expected to line up for Forest for the final time in Sunday’s play-off final against Huddersfield at Wembley.
He made 71 total appearances for Boro before moving on loan to Forest at the start of the season.
Spence is also an England Under-21 international, making his debut as a substitute during a 3-0 victory away to Albania on 29 March 2022

He would be behind Son and Kulusevski as a wide forward. Basically competing with Moura for a bench spot.
He is too expensive to be a back up when we also need to spend money on wing backs, defenders and creative midfielders.
Why is this argument not applied to Jota/Dias/Firmino/Origi?
WE NEED QUALITY DEPTH.
Signing someone like GJ would allow us to play Son in the CL, for example, and then rest him and start GJ in the league. These kind of options are what is required to go to the next level.
Some unique phenomenon that Spurs seem to have that we can never sign an attacker because Son or Kane couldn't possibly ever be rotated. The reason they have seen so little rotation is precisely because we never had a good option to come in, so they were forced to play every minute even through severe losses in form.
I’m not convinced by Gabriel Jesus, 28 appearances 8 goals. That’s poor playing for a team like Citeh.
It boils down to the player's motivations, chance of a trophy or the ability to buy-off a players desire to play every week.
It's not a Spurs exclusive issue.
Before they signed Jota and Diaz they fixed more urgent problems though ... goalie, centre back, central midfield.
